Randomized Phase 2 Study Of IMC-A12 And IMC-1121B For Advanced Prostate Cancer Commences Patient Enrollment

�ImClone Systems Incorporated (NASDAQ: IMCL), a global loss leader in the development and commercialization of novel antibodies to treat cancer, today announced that its disease-directed Phase 2 clinical trial in patients with advanced prostate cancer randomized to treatment with either IMC-A12 or IMC-1121B plus mitoxantrone and prednisone has commenced patient registration. IMC-A12 and IMC-1121B ar two therapeutic candidates in ImClone's proprietorship receptor-targeted antibody pipeline. IMC-A12 is ImClone's fully human, IgG1 anti-insulin-like growth factor-1 receptor (IGF-1R) monoclonal antibody and IMC-1121B is its fully human, IgG1 anti-vascular growth factor receptor-2 (VEGFR-2) monoclonal antibody.


This multicenter, randomized open-label Phase 2 single-arm study is enrolling patients with metastatic androgen-independent prostate cancer who have developed disease progression during or within 60 days of receiving docetaxel-based chemotherapy or demonstrated intolerance to docetaxel-based therapy. A sum of 132 patients are expected to be enrolled at various centers, including those that participate in the Department of Defense's Prostate Cancer Consortium. This Phase 2 study is designed to evaluate the efficacy and safety of both IMC-A12 and IMC-1121B combined with mitoxantrone and prednisone. IMC-A12 and IMC-1121B are administered weekly, whereas mitoxantrone is administered every three weeks with oral daily prednisone.


"We ar very pleased about beginning another disease-directed, proof-of-concept run which efficiently evaluates two of our pipeline antibodies with discrete mechanisms of action, still each with potential applicability in the treatment of prostate cancer based on preclinical investigations performed by ImClone and academic collaborators," said Eric K. Rowinsky, M.D., Chief Medical Officer and Executive Vice President of ImClone. "The results of whatsoever one of the study arms, or even both, in conjunction with those of other ongoing clinical and preclinical studies, may serve as platforms for subsequent registration directed bodily process in several early- and advanced-stage prostate gland cancer settings."


IMC-A12 is a fully human IgG1 monoclonal antibody designed to specifically target the human IGF-1R, thereby inhibiting certain ligands known as IGFs 1 and 2 from binding to and energizing the receptor. This action blocks a signaling nerve pathway that enhances tumor cell proliferation and survival. In 2007, ImClone completed enrollment into two Phase 1 studies of IMC-A12 which demonstrated friendly safety and pharmacokinetic profiles, as comfortably as prelim evidence of antitumor bodily process as a single federal agent when administered either weekly or every two weeks. In addition to this Phase 2 study proclaimed today, Phase 2 studies of IMC-A12 in adult and adolescent patients with soft tissue sarcoma, untreated advanced prostate gland, pancreatic, colorectal, liver, and head and neck cancers, as well as a series of Phase 1/2 studies in pediatric malignancies and another evaluating the combination of IMC-A12 and temsirolimus, have begun to enroll patients.


IMC-1121B is a fully human IgG1 monoclonal antibody designed to bind to the extracellular domain of VEGFR-2 launch on tumor vasculature, thereby inhibiting certain ligands known as vascular endothelial growth factors from binding to and activating the receptor. This natural action blocks a signaling pathway key to new blood vessel formation in growing tumors, which has been shown to starve tumors of their nutrient issue and solution in significant tumor growth inhibition in pre-clinical models. In addition to this Phase 2 study announced today, disease-directed studies of IMC-1121B in patients with advanced malignant melanoma, liver and renal cancers have begun to inscribe patients, and additional Phase 2 and 3 evaluations are in various stages of development. In April 2008, ImClone announced an agreement with the Food and Drug Administration on a Special Protocol Assessment for a Phase 3 study of IMC-1121B in women with metastatic chest cancer, which recently commenced.

Avril "Too Sexy" for Malaysia?

Avril Lavigne is getting under the skin of Malaysian censors.


A hard-line Islamic opposition party in the Southeast Asian country is calling on the politics to opening an upcoming concert featuring the "Sk8er Boi" isaac M. Singer, calling her stage pretend way "excessively sexy" for local tastes.

Frank Darabont’s LaBeouf-less ‘Indiana Jones 4’ Screenplay Leaks Online

The script for Indiana Jones and the Kingdom of the Crystal Skull wasn't born terrible — during its two-decade development, it went through multiple revisions and writers before George Lucas finally deemed it bad enough to shoot. And now, Frank Darabont's 2003 draft (called Indiana Jones and the City of the Gods) has hit the Internet! According to reports, Steven Spielberg loved it and Lucas hated it, which probably means it's fantastic. We haven't read it yet, but we skimmed it; Crystal Skull's killer ants are in there, and Indy, Marion, and Professor Oxley still implausibly survive a boat trip over four waterfalls (also — SPOILER ALERT! — knowledge is still their treasure). What's different? Among other things, Shia LaBeouf's character is nowhere to be found, which means nobody swings Tarzan-style through the jungle surrounded by CGI monkeys (CGI monkeys do make an appearance, but, on the page at least, they seem relatively non-annoying). Oh, what might've been!

Denise Richards - Richards Asks Sheen To Be Civil


DENISE RICHARDS has offered an olive branch to her ex-husband CHARLIE SHEEN - pleading with the star to stop publicly bashing her now he has re-married.

The former couple divorced in 2006 and has since been embroiled in a bitter dispute over custody of their two daughters, three-year-old Sam and Lola, two.

And they have taken any opportunity open to them to showcase their distaste for each other, with Sheen accusing the beauty of sending him emails begging him to donate his sperm so that she could carry another one of his children.

The claim infuriated Richards, who slammed the star, saying, "I don't want Charlie's prostitute-tranny-infested sperm", when defending herself to New York's gossip column Page Six.

Sheen also blasted their time as husband and wife after his recent marriage to Brooke Mueller, saying, "It feels like my first real marriage (to Mueller). The first one (to Donna Peele) was a show, the second (to Richards) was a con, and this one is the real deal."

But, now, Richards claims she is tired of their public feuding, and is hoping her former partner will act "civil" towards her now that he has a new wife.

She says, "He loves to trash me in the press and discredit me and has tried to sabotage my show. I can't change that, so I just keep living my life.

"I hope that now that he's married, he can move forward and be civil."

Jaws star Roy Scheider dies, aged 75

Roy Scheider, the actor best known for his role as the beleaguered police chief Martin Brody in 'Jaws', has died. He was 75.
The two-time Oscar nominee died of complications from multiple myeloma, a cancer of the blood cells, at the Myeloma Institute for Research and Therapy at the University of Arkansas for Medical Sciences in Little Rock.
Hospital spokesperson Leslie Taylor said the actor had been treated there for the disease at different times over the past two years.
New Jersey-born Scheider earned his first Oscar nomination (Best Supporting Actor) in 1971 for 'The French Connection'.
Eight years later he was nominated for the Best Actor Oscar for his performance in 'All That Jazz'.
Among his other credits were 'Klute', 'Marathon Man', 'Still of the Night' and the TV adventure series 'SeaQuest DSV'.

Jennifer Lopez welcomes twin babies

Jennifer Lopez and her husband Marc Anthony are celebrating the birth of twin babies in New York.
According to People magazine, the babies, a girl weighing 5lbs 7oz and a boy weighing 6lbs, were born early this morning.
Lopez's manager Simon Fields confirmed the news to the magazine, saying: "Jennifer and Marc are delighted, thrilled and over the moon."
The twins are the first children for the couple, who married in June 2004. Anthony already has three children.

EastEnders star attacked leaving nightclub

'EastEnders' star Lacey Turner returned to filming in London today after getting caught up in a fight when she left a nightclub.
The 19-year-old actress, who plays love cheat Stacey Branning in the hit BBC1 soap, was on her way home from a night out with co-star Aaron Sidwell (Steven Beale), also 19, when she was hit.
Turner did not report the incident, which took place at 2am at a supermarket car park near the Destiny club in Watford, Hertfordshire.
The actress' spokeswoman said: "She was literally walking back into a car park when a fight broke out and she got clipped.
"She was in the wrong place at the wrong time but she's fine. She and Aaron were not involved in the fight.
"She had a day off yesterday because she was off schedule and she's back filming today."
Turner joined the soap in November 2004. Her feisty character has recently been searching for her husband Bradley after he disappeared when he found out she had been cheating on him with his father.

Bailey Rae's husband is found dead

The husband of British soul singer Corinne Bailey Rae was found dead in Leeds on Saturday from a suspected drugs overdose.
West Yorkshire police said that they were called to an apartment in the Hyde Park area of the city where they found the body of a 31-year-old man, later identified as Jason Rae.
Police said that a 32-year-old man was arrested under suspicion of supplying drugs and freed on bail.
A post-mortem on the body of Jason Rae, a saxophonist with the group the Haggis Horns, proved inconclusive, and police are awaiting toxicology reports.
In a statement, Bailey Rae's record label EMI said: "EMI Records would like to offer its sincere condolences to Corinne Bailey Rae and the Rae family at this tragic time. We ask that the media respects Corinne's privacy and that of her entire family."
